The airport sits at Yalyalup, east of Busselton in Western Australia's South West, about 220 km south of Perth. It opened on 15 March 1997 and replaced the wartime Busselton Aerodrome, which dated from 1940. Margaret River was added to the name in 2015, when the airport was known as Busselton Regional Airport.

It has one runway, 03/21, rated for Code E aircraft. A redevelopment that began in 2017 cost A$69.7 million. It lengthened and strengthened the runway and added a Code E apron, a general aviation precinct, new lighting and more parking. A Royal Flying Doctor Service facility opened in June 2024.

Passenger traffic is split between scheduled flights and fly-in fly-out charters. Rio Tinto began fly-in fly-out flights from the airport in 2007, and charter operators still fly workers to Pilbara mine sites from here. Jetstar began a three-year trial of Melbourne flights in April 2022 and added Sydney in March 2024. About 156,000 passengers used the airport in 2024, up from roughly 25,000 a year before COVID-19. A planned new terminal was put on hold in 2018, so the upgraded 2014 building remains in use.
