Kirksville Regional Airport is a city-owned public airport about 6 km south of Kirksville, Missouri, on the west side of U.S. Highway 63. The federal government built the site in 1930 as Intermediate Field, Site 12, on the transcontinental airway. An earlier airfield had opened on the north edge of town in 1924.

Scheduled airline service began in 1960 with Ozark Air Lines. Air Midwest, Air Choice One and Cape Air followed. Contour Airlines replaced Cape Air in 2023 and brought the first scheduled jet service to the field. Contour flies under the federal Essential Air Service program, which subsidizes the route. Twelve nonstop round trips a week run to Chicago O'Hare and Dallas/Fort Worth, with a contract through July 2029. Contour has interline agreements with Alaska, American, JetBlue and United.

Runway 18/36 is concrete and measures 6,005 by 100 feet. A second runway, 9/27, is turf and measures 1,370 by 100 feet. The FAA counted 5,888 aircraft operations in the year ending April 2023. Half were general aviation, 37 percent airline and 12 percent air taxi, and 20 aircraft were based on the field. Enplanements reached 5,375 in 2025. A new 6,500 square foot passenger terminal opened in June 2026 and gives passengers nearly twice the space of the old building.
